Well I don't think there's going to be anything new in GHIII besides new songs which will probably have exact custom song copies for PS2 GHII within two weeks.
However, if GHIII comes with online multiplayer, that would own so much.
Besides the already mentioned online play, the controllers handle better, and the HOPO engine works differently.

The wii controller fit in is awesome. We get vibrating guitars from the controller and the speakers on the controller for awesome sound.

I play guitar lefty and I play guitar hero righty. All the tapping from righty GH has made my lefty plucking skills on guitar faster. It has however made my lefty picking feel odd, but I don't care. I much rather prefer plucking anyway. Especially on bass.

No man, just turn on lefty-flip, it'll flip the buttons around so that Green is on the right and Orange is on the left; aka the equivalent of a lefty guitar
